Famiglia originaria da Peretola nei pressi di Firenze. I V., immatricolati nei tempi più antichi tra i vinattieri e poi tra i giuridici e notai, dettero alla Repubblica 25 priori e 3 gonfalonieri di giustizia dal 1450 al 1524; e per le nobili parentele costantemente contratte furono più volte ricevuti per giustizia nell'Ordine di Malta, nel quale Niccolò, di Simone, nel 1495, fu fatto commendatore, e in quello di S. Stefano di Toscana. Ad essi si deve la fondazione dell'ospedale di S. Maria dell'Umiltà, detto poi di S. Giovanni di Dio, fatto costruire sulle loro case di Borgognissanti. Furono ascritti al patriziato fiorentino con decreto del 13 marzo 1752. Questa famiglia universalmente nota per aver dato in natali al celebre navigatore Vespucci Amerigo, navigatore fiorentino (1454-1512), prima addetto alle imprese commerciali dei Medici, poi al servizio della Spagna.
