Casata assai antica e nobile, cospicua in Alessandria, Assisi, Bologna ed altre città fin dal 1200, propagatasi, nel corso dei secoli, in diverse regioni d'Italia. La cognominizzazione è anteriore al XII secolo, si vuole sia una patronimia derivata dal nome medioevale di origine germanica Ghislerio. Nei ricordi dei tempi andati si leggono alcune varianti di questo cognome per le solite cause della fonetica storica e degli errori di trascrizione; è comune opinione tra gli studiosi di araldica che Ghisleri, Ghislerio, Ghisileri etc. siano tutti rami di un unico ceppo originario. In Assisi nel 1200 la famiglia è annoverata tra i nobili di quella città. In Bologna esiste almeno dal 1200, una delle torri della città è appunto la torre dei Ghisilieri, una delle cinque torri del quartiere di Porta Stiera appunto possedute da questa famiglia gentilizia che aveva radici anche a Pistoia, famoso fu il capitano pistoiese Grandonio
