Antica ed illustre famiglia, originaria di Matera, le cui prime vestigia risalgono al 1198, anno in cui viveva Eustachio Mantoro, barone di Cancellara, Castelnuovo e Casale di S.M. di Giambove. Lo storico materano Gattini cita, poi, altri atti notarili nei quali, dopo una lacuna di circa un secolo e mezzo, si leggono i nomi di altri componenti della famiglia: nel 1402, in un atto del notaio Tuccio de Rahone, è citato Stefano, di Giovanni de Santoro; nel 1432, in un atto del notaio Giovannuzzo de Berardo, si legge il nome di Nicola Giovanni di Santoro; nel 1476 risulta sindaco di Matera Toto Santoro, dal quale nacquero Pietro e Lorenzo, dei quali quest'ultimo sposò Caterina de Angelis, da cui ebbe quattro figli: Vincenza che sposò un Del Duce; Laura che sposò Alfonso Mantoia; Giustina coniugata con Leonardo d'Aurilia ed infine Agostino, che divenne sacerdote.
