La famiglia è originaria di Corsignano, che poi Pio II denominò Pienza, ma venne presto in Siena, dove nel 1147 costruì un sontuoso palazzo, nel quale alloggiò, fra gli altri, l'imperatore d'Oriente, Emanuele. Più tardi i Marsili furono ascritti al Monte dei Nove e Leonardo, che già aveva partecipato alle Crociate, si distinse alla battaglia di Montaperti. Già al tempo del pontificato di Bonifazio VIII questa casata ebbe un vescovo nella persona di fra Camillo, nominato da quel papa alla sede di Otranto. Nel 1333 troviamo nel supremo magistrato di Siena, Piccolo, di Scotto. La famiglia possedeva in Maremma il castello della Marsigliana, da cui forse presero il nome. Comperarono nel 1345, dai conti Aldobrandeschi, coi quali erano uniti da vincoli di parentela, la signoria di Collecchio, per la quale offrivano ogni anno un cero alla cattedrale di Siena nella festa dell'Assunzione.
