La famiglia è originaria della Val di Chiana e, dall'appellativo di ser che si trova ai suoi più antichi rappresentanti, appare che esercitasse la professione di notariato. Il primo che troviamo in Siena è ser Neri di ser Giovanni, che nel 1392 prese parte al governo del Comune, e nel 1405 fu eletto Capitano del Popolo. Al governo di Siena parteciparono anche il figlio e il nipote di Neri: Giovanni e Neri, pur mantenendo la dimora in Foiano, dove il figlio dell'ultimo, Rosado, edificò nel 1515 una cappella gentilizia nella collegata. Neri e Luca di Rosado furono incaricati di ricedere e tradurre in italiano gli statuti di Foiano nell'anno 1539. Un altro discendente della famiglia, Niccolò di Neri, rivolse supplica alla Signoria di Siena verso la metà del 600, perchè gli fosse riconosciuta la cittadinanza e nobiltà senese ed ottenne simile dichiarazione dalla quale appare che i Serneri appartenevano al
