Antica ed assai nobile famiglia oriunda da Cana, nella Maremma senese, i cui possedimenti però si estendevano fino alla Valle della Chiana, propagatasi, nel corso dei secoli, in diverse regioni d'Italia. Trasferitasi a Siena, sin da epoche remote, vi esercitò l'arte dei panni di lino, da cui le venne il cognome. Durante la guerra aragonese contro la Francia del sec. XV Simone Pannilini sostenne i primi inviando ingenti quantità di truppe e danaro a Ferdinando I di Napoli, il quale apprezzandone il gesto convertì in feudi i possedimenti del Pannilini nominandolo, insieme al fratello Mino ed alla sua discendenza tutta, Patrizio di Siena. I Pannilini risultano iscritti nel Monte del Popolo, creato verso il 1360, ma il primo di essi che abbia partecipato effettivamente al governo di Siena fu Pietro, di Mino, di Paolo, nel 1429. Nanni, di Barna, fu provveditore di Biccherna nel 1432, e già nel 1403 era
