Antica famiglia napoletana di cui, si ha notizia fin dal tempo della dominazione normanna, e che riconosce a suo capostipite un Roberto de Regina, feudatario di re Guglielmo II, il Buono. Ricevuta nel S. M. O. di Malta, in Priorato, nel 1664, come quarto materno del cavaliere Giacomo d'Aquino; e nel 1702, quarto ava materna del cavaliere Gio. Battista Cioffi, avendone fatta la prova da Orso di Regina, primo stipite, dell'anno 1322. Fra i personaggi illustri di questa casata si ricordano Nicola Antonio, creato cardinale da papa Giulio II; Giovanni Antonio, giustiziere di Lecce nel 1577; Francesco, giustiziere di Calabria nel 1596. I de Regina ebbero in feudo il castello di Macchia di Valfortone, antecedentemente al 1482, poichè, in quell'anno, Francesco de Regina ne ebbe rinnovata l'investitura da re Ferdinando I di Aragona.
