Antica ed assai nobile famiglia portoghese, sulla cui origine vi sono diverse ipotesi da parte dei genealogisti. La tradizione vuole che suo capostipite sia un Vaso Pires de Sampaio, signore di Vila Flor Chacim, Ansiães, Vilarinho da Castanheira e altri feudi in Trás-os-Montes, fiorente ai tempi di Re Don Fernando e Giovanni I del Portogallo. Alcuni genealogisti pretendono che questo Vasco Pires de Sampaio fosse quel Vasco Pires Osório, figlio di D. Pedro Álvares Osório, conte e marchese di Trastamana e signore di Casa de Vilalobos, e di sua moglie D. Brites de Catros, oriundi dalla Galizia, i quali rifugiarono in Torre de Moncorvo in seguito all'omicidio di un gentiluomo galiziano. Altri autori affermano, poi, che sotto il regno di Dionigi del Portogallo, fioriva un Martim de Sampaio, signore di Sampaio e discendente del Conte D. Ermenegildo, avo di Vasco Pires de Sampaio.
