Antica ed illustre famiglia siciliana, originaria di Adernò. Vuole la tradizione che fosse originaria dalla Spagna, ove risiedeva sotto altro nome e che il capo di essa, prendendo parte alla prima Crociata per la conquista di Terra Santa, assumesse il nome di Spedalieri. Comunque un Antonio Spedalieri, regio milite nel 1465, e dimorante a Palermo, ottenne dal re Giovanni d'Aragona il permesso, insieme ai Gioeni, di armare a proprie spese delle triremi per combattere i Saraceni. Suo figlio Giovan Vincenzo fu vicario generale del regno e nel 1500 passò con la fam. ad Adernò come governatore.
